7th C1 Inhibitor Deficiency Workshop, Budapest 20-22 May 2011

Colleagues, students, and representatives of pharmaceutical companies involved in the therapy or research of C1-inhibitor deficiency and hereditary angioedema are expected to attend from all over the world. Patients with hereditary angioedema, who wish to know or present novelties and new results in this area are also welcome. Similar to the previous six meetings held in Hungary, the aim of this conference is to enhance clinical research and pharmaceutical development by relying on the latest results, and to help the global introduction of standardized diagnostic and therapeutic methods.

Berinert P in Romania

In 2009, CSL Behring GmbH, Germany, has been granted authorization to market Berinert P in Romania for the treatment of acute hereditary angioedema attacks. Berinert P is a purified, virus-inactivated, human plasma-derived C1 inhibitor concentrate. Berinert is now licensed in 28 countries around the world including many European countries. CSL Behring completed the European Mutual Recognition Procedure (MRP) for Berinert in 23 countries in December, 2008. Marketing authorization was also granted for USA in november 2009 and Australia in January, 2010.

Firazyr for the Romanian HAE patients

The European Commission has granted in 2008 Jerini AG marketing authorization for its compound Firazyr (R) (Icatibant) in the treatment of acute attacks of hereditary angioedema in all EU countries. Firazyr works by blocking the B2 receptor as an antagonist to the peptide-hormone bradykinin. Bradykinin has been shown to be elevated in HAE patients and responsible for edema formation during HAE attacks.

Frequency, timing, and type of prodromal symptoms associated with hereditary angioedema attacks

According to a study recently published in Allergy and Asthma Proceedings, 87.0% of HAE patients report the presence of prodromal symptoms before their last HAE attack and 95.7% pf them reported having had prodromal symptoms before HAE attacks in the past. The most commonly reported prodromal symptoms included unusual fatigue, rash, and muscle aches. Prodromes occur frequently before HAE attacks.

Novel mutagenesis mechanism for hereditary angioedema pathogenesis

In a recent study done by a group of Greek researchers 3 unrelated families with recurrent angioedema attacks and decreased C1 inhibitor antigenic levels were analyzed for SERPING1 mutations. One family displayed a novel SERPING1 alteration, characterized by the substitution of two consecutive nucleotides TC to AA, resulting in a termination codon (F225X). The authors consider that this is the first report of such a mutation in SERPING1, causing HAE. The second and the third family displayed mutations already described in the literature.
